Keratinocyte differentiation is characterized by expression of differentiation markers and cellular morphological changes. Keratin 10 (K10) is marker of keratinocyte differentiation, correlating directly with the progress of differentiation. K10 expression triggered by photodynamic treatment mediated by aluminium phtalocyanine disulfonate were followed in primary human epidermal keratinocytes HEKa. Analysis of total cell population may hide subgroup of differentiated keratinocytes. Cellular imaging plays a significant role in subtyping of cell population. The aim of the study was to perform immunofluorescence microscopy and using biostatistical modeling to identify subgroups based on the fluorescence level of K10 in HEKa. Data analysis identified two subgroups of K10 positive cells. Procedure of biostatistical modeling can be extended and applied to bioinformatics tools for cell imaging.
