Adenosine triphosphate binding cassette transporter 1 (ABCA1) has been previously shown to be the major element controlling cerebral malaria susceptibility in Plasmodium berghei ANKA infected mice. The advancement of human genome project and molecular technology facilitates the analysis of human molecular mechanism responsible for severe and cerebral malaria. To examine whether human genetic polymorphisms in ABCA1 gene influence on the susceptibility to severe malaria and cerebral malaria in Thai patients infected with Plasmodium falciparum. We examined the possible association of sixteen single nucleotide polymorphisms (SNP) spanning a 150 kb region of the ABCA1 gene with malaria severity in 477 malaria patients living in the northwest region of Thailand. The ABCA1 SNP (-402G/G) located in the putative promoter region was significantly associated with susceptibility to severe malaria.
