The Electron Microscopy Data Bank is the international repository for high-resolution electron microscopy density maps. It has become a standard resource for structural biology now operated jointly between the EBI and the RCSB at Rutgers University in New Jersey. In the future, cryo-electron tomography is expected to provide a bridge between structural, cellular, and systems biology in trying to understand macromolecular complexes as molecular machines in a cellular context. To adapt to the growth in image data from such technologies, we are developing tools to represent entries that contain "rich content", including 3D density maps, atomic structure coordinates, averaged submaps for resolution improvement, ensembles of maps resulting from classification, and markup information such as segmentation masks. A standardized flexible representation based on content graphs (RDF) is the basis for integration of computational analysis and visualization tools. Ontologies can be used to make even diverse datasets browsable to the user, while web-based visualization, web services, and grid/cloud computing support support a rich variety of applications on the database.
